Why Proper Cleanroom Cleaning Matters

Cleanrooms depend on both engineering controls and disciplined operations. Whether the space supports healthcare, biotechnology, pharmaceuticals, aerospace or another contamination-sensitive process, an effective cleaning program helps control particles, residues and, where relevant, microorganisms on surfaces.

The cleaning program should be defined in written procedures based on the room's use, contamination risks, surface materials, applicable regulations, and quality-system requirements. Inadequate cleaning can contribute to environmental excursions, product or process contamination, investigations, corrective action, and operational delays.

ISO 14644-1 classifies air cleanliness by airborne particle concentration, while ISO 14644-5:2025 addresses the broader operations control program, including cleaning. Pharmaceutical, compounding, healthcare, and other facilities may also have industry-specific cleaning and disinfection requirements. Cleaning procedures should therefore be developed for the actual process rather than copied from the ISO class alone. The facility should define what is cleaned, by whom, with which materials and agents, in what sequence and at what frequency, along with required contact times, residue-removal steps, documentation and response to an unacceptable result.

How To Develop and Implement a Cleanroom Cleaning Procedure

  1. Preparation 
    • Gather Supplies: Assemble the supplies identified in the facility's procedure. Depending on the application, these may include:

      • Cleaning agents, disinfectants and sporicidal agents selected for their intended purpose and demonstrated compatibility

      • Low-lint or non-shedding wipes, mop heads and applicators; sterile supplies where required

      • Cleanroom-compatible HEPA-filtered vacuum equipment, if permitted by the procedure  

      • The gowning and personal protective equipment specified for the room, task and chemicals used

      • Dedicated tools and supplies assigned to the appropriate room or cleanliness zone  

      • Buckets, wringers or closed cleaning systems constructed of cleanable, compatible materials  

        • Review Protocols: Ensure you're familiar with cleanroom cleaning protocols and procedures. These may vary depending on the class of cleanroom and the specific requirements of your facility.

        • Personal Preparation: Wear appropriate cleanroom attire, including gowns, gloves, masks, and shoe covers. Ensure your attire is free of any contaminants. 

  2. Initial Cleaning 
    • Remove Waste and Unnecessary Items: Remove waste and items authorized for removal without carrying contamination into cleaner areas. Materials that remain in the room should be handled or protected as specified by the procedure.
      • Remove Loose Contamination: Where permitted, use an approved HEPA-filtered vacuum or damp method that minimizes particle redistribution. Follow the facility's defined sequence, generally moving from cleaner to less-clean areas, from higher to lower surfaces and from the back of the room toward the exit.
      • Clean and, Where Required, Disinfect Surfaces: Apply the specified agent using the approved wiping technique. Use overlapping strokes and change wipes or applicator surfaces often enough to avoid spreading contamination. Keep surfaces wet for the required contact time and follow the approved procedure for rinsing or residue removal.
  3. Floor Cleaning 

    • Remove Loose Particles: If the procedure includes an initial particulate-removal step, use a dedicated low-lint mop, approved HEPA-filtered vacuum or damp method. Conventional dry sweeping or dusting can redistribute particles and should be avoided.
    • Wet Mopping: Prepare or dispense the approved solution as directed. Use dedicated cleanroom tools and a method that separates clean solution from recovered soil where required. Mop with overlapping passes from the cleaner or farthest area toward the exit without walking over completed sections.
    • Rinsing and Drying: Perform any required rinse or residue-removal step using the specified water quality or solution. Allow the surface to dry or complete the validated wet-contact time before the room returns to use, as directed by the procedure.
  4. Equipment and Tool Cleaning 

    • Prepare Equipment: Place equipment in a safe state and disassemble only as authorized by the manufacturer and facility procedure. Protect sensitive components and maintain any required line clearance or material controls.
      • Clean and Disinfect: Use agents and methods compatible with the equipment and process. Give particular attention to product-contact areas, touch points, joints, and other difficult-to-clean locations while avoiding damage to sensors, finishes or airflow components.
      • Reassemble and Inspect: Reassemble the equipment as instructed, inspect for cleanliness and residue, and complete any required operational check, release or line-clearance documentation before use.
  5. Final Steps 

    • Inspect the Cleanroom: Check the defined areas for missed surfaces, visible soil, damage, standing liquid and chemical residue. Escalate any condition that cannot be corrected within the approved procedure.
      • Document the Cleaning: Record the date and time, room or equipment, activity performed, agents or lot numbers where required, personnel, completion status and any deviation or unusual condition. Documentation should meet the facility's data-integrity and record-retention requirements.
      • Manage Supplies: Dispose of single-use materials as directed, segregate used reusable tools and restock supplies without compromising their status. Store agents and tools in their designated locations and within applicable expiration or use periods.
      • Return the Room to Service: Follow the facility's established recovery or release criteria before operations resume. Depending on the process, this may include drying, completion of the HVAC recovery period, inspection, or environmental checks rather than an arbitrary settling time.
  6. Regular Maintenance 

    • Establish a Schedule: Define frequencies by surface and activity based on process risk, room use, applicable requirements, environmental-monitoring trends and events such as spills, maintenance or excursions. Review the schedule when conditions or risks change.
    • Maintain Training and Qualification: Train personnel before independent work and reassess competency at the frequency required by the facility's program. Update training when procedures, agents, equipment, or applicable requirements change.
    • Verify Effectiveness: Periodically review records, observations, deviations, environmental-monitoring data and surface-condition trends. Use the results to investigate recurring issues and improve the procedure, training, agents, or frequencies where needed.
